Sequential IgE-epitopes of ovomucoid exposed by glutathione are relevant in persistent egg allergy

چکیده
Methods Reduced ovomucoid in raw eggwhite was detected using a fluorescence-labeled alkylation probe. The common antioxidants glutathione and cystein were used for in vitro linearization and effects were monitored by CD-spectrometry. Egg-allergic patients were tested serologically (n=19) and by skin prick test (n=9) for IgE against native and linearized ovomucoid and optionally its cooked state.

Molecular diagnosis of egg allergy.
PURPOSE OF REVIEW Allergy to hen's egg is common in infancy and childhood. Oral food challenges are often required to diagnose egg allergy, because of the limitation in the diagnostic accuracy of skin test and specific IgE to egg white. New molecular diagnostic technologies have been recently introduced into allergological research. In this article, we will review the recent literature regardin...
متن کاملUnscrambling Egg Allergy: The Diagnostic Value of Specific IgE Concentrations and Skin Prick Tests for Ovomucoid and Egg White in the Management of Children with Hen's Egg Allergy
Resolution of egg allergy occurs in the majority of egg allergic children. Positive specific IgE antibodies to ovomucoid (OVM) have been suggested to be of greater predictive value for persistent egg allergy than specific IgE to egg white. The performance of OVM-specific IgE antibody levels in a cohort of children referred for a routine egg challenge was compared with egg white specific IgE lev...
متن کاملMolecular diagnosis of egg allergy: an update.
Hen's egg allergy affects up to 2.5% of young children and is potentially life-threatening. Several phenotypes of egg allergy have been identified, including those who tolerate extensively heated egg in bakery products. Diagnosis and monitoring for resolution often requires oral food challenges, which can result in anaphylaxis. Newer approaches, such as component-resolved diagnostics, microarra...
متن کاملIdentification of IgE-Binding Peptides Derived from Chemical and Enzymatic Cleavage of Ovomucoid (Gal d 1)
Ovomucoid (OM), a major allergen of hen's egg white, consists of three trypsin inhibitory domains comprising 186 amino acids. In order to identify IgE-binding sites of its protein backbone OM was cleaved chemically and enzymatically. In SDS-PAGE/immunoblot experiments 10 peptides showing IgE-binding from human sera of egg allergic individuals were identified by amino acid sequencing. Up to five...
متن کاملThe Major Allergen from Hen's Egg White: Ovomucoid (Gal d 1)
Ovomucoid (OM) is the dominant allergen from hen's egg white. Frequency of sensitization to OM varies from 34-97% in egg allergic patients. The carbohydrate moieties of OM present up to 22-29%. It consists of 186 amino acids forming three tandem domains, each providing trypsin inhibitory activity. Effective isolation and purification of OM was attained by combined IEC / SEC and by automated fra...
